甘 油 三 酯(油脂、食品)酶法测定试剂盒 E1014

E1014105 assays750元描述:高油脂食品材料,如液态油脂、油料植物组织和种子、脂肪组织、肌肉间脂肪等的甘油三酯测定远比与血液甘油三酯测定复杂。试剂盒避免了有毒的氯仿甲醇提取、繁杂的氮气吹干和脂质复溶等步骤,采用高效能试剂进行裂解和甘油三酯抽提,以及优化的GPO Trinder酶学反应组分和操作步骤。简单易行,灵敏度高,线性范围20~2000 µmol/L。适用于农业、畜牧业、食品材料,用于动物脂肪肌肉、植物叶片种子、土壤、以及固态食品材料、含高浓度油料样品中甘油三酯的精确测量。

适于测定样品
(1) 动物脂肪组织、肌间脂肪
(2) 油料植物组织和种子如花生大米等
(3) 固态食品材料、土壤
(4) 液态成品油或含高浓度油脂的样品
其它类型的组织细胞甘油三酯测定用E1013;血液、培养液甘油三酯测定使用E1003。

测定原理:根据经典GPO Trinder反应,脂肪酶分解血甘油三酯为甘油。甘油激酶催化甘油生成3-磷酸甘油,后者被磷酸甘油氧化酶氧化生成过氧化氢,在过氧化物酶催化下进行生色反应,在550nm有最大吸收峰,光密度值与甘油三酯浓度成正比。

组成 (105次微板测定或30次1 ml比色杯测定)
(1) 裂解液 100 ml
(2) R1试剂 16 ml
(3) R2试剂 4 ml
(4) 4 mmol/L甘油标准品1 ml
4 ºC避光储存3月。

所需设备:酶标仪、生化分析仪或721、722型可见光分光光度计。96孔微板或比色杯光径1cm。最佳工作波长550nm,如无此波长建议优先选用570nm、次选530、490nm。
